Current studies show that Peyronie's disease (hardening of the penis) is an inflammatory disease of the penis in which the elastic layer (tunica albuginea) of the penis in the central tissue (corpora cavernosa) is damaged. As a result, the affected area loses elasticity and when it fills with blood it does not stretch, but causes the penis to curve in the direction of the scar. Peyronie's disease is the most common cause of excessive curvature of the penis.

The process begins with the oxidation of various fats and protein molecules that lead to inflammation of the elastic layer (tunica albuginea) and corpora cavernosa. This continues until finally the collagen fibers are replaced by non-elastic fibers that form the fibrous plaque (hardened nodules) very characteristic of this disease.

These plaques can be felt under the skin, usually on the top or sometimes on the sides of the penis. Most of them dissolve on their own naturally, but sometimes they persist and over time cause:

Abnormal curvature of the penis (often upwards);
